Expression of Autophagy and Mitophagy Markers in Breast Cancer Tissues

Mitochondria play important roles in regulating cell bioenergetics status and reactive oxygen species (ROS) generation. ROS-induced mitochondrial damage is among the main intracellular signal inducers of autophagy. Autophagy a cellular catabolic process that regulates protein organelle turnover, while selective form autophagy, mitophagy, specifically targets dysfunctional degradation. This study aims to measure levels oxidative stress, apoptosis invasive breast carcinoma tissues using immunohistochemistry (IHC). Tissue microarrays 76 patients with cancer were stained six IHC markers (MnSOD, Beclin-1, LC3, BNIP3, Parkin, cleaved caspase 3). The expression intensity was determined for each tumor tissue adjacent tumor-matched control tissues. Intermediate strong staining scores MnSOD, LC-3, BNIP-3, Parkin significantly higher compared matched control. scoring further classified into negative positive staining, which showed Beclin-1 high other markers. Positive association also noted between BNIP-3 as well LC-3 caspase-3 immunostaining. To our knowledge, this one first studies both mitophagy autophagy same findings from will be great potential identifying new biomarkers inspire significant interest applying anti-autophagy therapies possible treatment cancer.
